They must wear white coats to prevent microbes from their clothes contaminating the samples. The white coat must be taken off before they leave the lab.

In a special jelly called agar is used.

A
All the necessary nutrients required for the bacteria to grow.

How is agar used in Petri dishes
A
It is warmed to make it sterile and then poured into Petri dishes

A
This creates a convection current to stop bacteria from the air falling into the samples.
