• Inhibition of COX pathways = ↓ prostanoid synthesis
- ↓ TXA2 preventing platelet aggregation
- ↓ PGI2 antithrombotic activity

- GIT Bleeding
- Peptic Ulcer
- Pseudoallergy reactions
Thienopyridine drugs?
Ticlodipine, Clopidogrel
Thienopyridine Mechanism of Action?
• Inhibition of platelet aggregation by antagonism of ADP receptors
- P2Y1: change in platelet conformation
- P2Y12: ↓ cAMP, ↓ ADP
Thienopyridine Side Effects and Contraindications?
- Well tolerated
* Mild nausea, dyspepsia, rashes
GPIIb/IIIa Antagonists drugs?
Abciximab
GPIIb/IIIa Antagonists Mechanism of Action?
• Antagonism of GPIIb/IIIa receptor and therefore prevent
platelet aggregation and activation
GPIIb/IIIa Antagonists Side Effects and Contraindications?
- Bleeding, Thrombocytopenia
* Allergic reaction (to mouse component)

Heparin Mechanism of Action?
- Binds to antithrombin and increases its activity in cleaving clotting factors (2, 7, 9, 10)
- Increased TFPI release
- Antidote = protamine sulphate
Heparin Side Effects and Contraindications?
- Increased bleeding tendency
- Allergic reaction (switch to synthetic heparin)
- Heparin induced thrombocytopenia (HIT)
- Osteoporosis and increased fracture risk
- Reversible alopecia, mineralocorticoid deficiency
