- Adrenergic antagonist
Chlorpromazine side effects
- Decrease the seizure threshold
- Alpha-adrenergic antagonisty = hypotension
- Anticholinergic effects
- Potentiate CNS depressant effects of other drugs
- Hyperprolactinemia
- Neuroleptic malignant syndrome
Torsades List Drugs
Some 1st generation antipsychotics come with a risk of Torsades de Pointe and can extend the QT interval
Acute dystonias (EPS)
involuntary movements and spasms of the face, neck, and tongue which make swallowing difficult
Akathasia (EPS)
Subjective feeling of wanting to be constantly in motion. Treat w/ anticholinergics or antianxiety drugs
Pseudoparkonism (EPS)
tremor, muscle twitch, and rigidity. Treat w/ anticholinergics
Tardive dyskinesia
Occurs after > 1 year of treatment
Involuntary movements of the facial muscles
5-HT1a location and MOA
Postsynaptic neurons in which agonists enhance dopamine in cortex
5-HT2a location and MOA
Presynaptic neurons and antagonists enhance dopamine in cortex
Major side effects of 2nd generation antipsychotics
- Weight gain
- Diabetes mellitus
- Cholesterol levels
Clozapine adverse effect
Increased incidence of agranulocytosis.
Monitor of leukocyte count
