intereferes with DNA and RNA polymerase synthesis

- competes with hypoxanthine & guanine for the enzyme HGPRTase & is convereted to thioinosinic acid
- inhibits DNA synthesis

leukemia, Non-Hodgkins, psoriatic arthritis, polycythemia vera, Crohn’s

Vinka alkaloids
MoA for Vincristine
binds to tubulin dimers, inhibiting assembly of microtubule structures & arresting mitosis in metaphase (inhibiting DNA synthesis)
Indications for Vincristine
non-Hodgkins, ALL
Drug class for Paclitaxel
-Taxanes
MoA for Paclitaxel
-Inhibiting assembly of microtubule structures, arresting chromosome segregation & cell division
–inhibits DNA synthesis by inhibiting formation of the mitotic spindle in the “M” phase
Indications for Paclitaxel
-ovarian, breast, lung, bladder, prostate, melanoma, esophageal, Kaposi’s sarcoma
*not leukemias & lymphomas

- interacts w/ DNA binding macromolecular biosynthesis
- inhibits topoisomerase II
- unclear bu inhibits DNA synthesis
