Diazepam (Benzodiazepine)
facilitate GABA at GABA-A R. muscle relaxant. increase inhibition 1° afferents in SC, enhances hyperpolarization. treat chronic spasm. oral or parental admin
Baclofen
agonist at GABA-B R. muscle relaxant. orally active, t1/2 3-4 hrs, hyperpolarization, facilitates inhibition spinal αMN, inhibits release substance P, reduce pain. less sedation than diazepam. treat severe spasticity
Dantrolene
inhibit Ca++ release from SR. direct acting muscle relaxant, binds to ryanodine R in SR. oral admin. but inefficiently absorbed. t1/2 8 hrs. treat malignant hyperthermia. Adverse weakness, sedation, hepatitis
Botulinum Toxin
inhibits presynaptic ACh release. flaccid paralysis. treat spastic disorders, lasts weeks-months. treat strabismus and much more importantly, wrinkles
