Q
Steps of looking at cells in the body, e.g. liver cells.
A
- Remove from body
- Fixation- preventing them from naturally degrading and damage due to processing for microscopy, preventing autolysis (self death). This often uses temperature, acids, microwaves, and chemicals
- Mount on medium for sectioning (often using paraffin wax block) and trimmed
- Cut using a microtome sliding into 5-7 micro metre thick
- sections of paraffin then floated on warm water bath and picked up onto glass slides, dried, wax removed, rehydrated and stained with histological stains
- Cover with coverslip to prevent from drying out

Q
can cell membranes be seen using a light microscope?
A
no
