3 important parameters to consider when coiling of sputum sample
Thickness
Evenness
Size
Size of coiled sputum smear
2 cm x 3 cm
It is used to stain bacteria that have high lipid content in their cell wall
Acid fast stain
Where does the primary stain binds in the cell wall
Mycolic acid
Color of Acid fast Bacilli & from what dye?
They are stained Red from carbol fuschin
Color of Non-Acid fast bacili and what dye is used?
Colored as blue or green from methylene blue or malachite green
Uses steam as a mordant
Ziehl-neelsen method/hot staining method
Same reagent but higher concentration of phenol w/c serve as a chemical mordant; sometimes have a wetting agent tergitol to fasten up reaction
Kinyoun’s/cold staining method
It differentiates M. Smegmatis to M. Tuberculosis
Pappenheim’s method
Differentiates m. Leprae from m. Tuberculosis
Baumgarten’s method
