Afb Staining Flashcards
TB is caused by a bacterium called?
Mycobacterium tuberculosis
M. tuberculosis usually attacks:
Kidney, spine & brain
Two tb related condition exist:
Latent TB Infection
TB Disease
How to diagnose TB disease?
By patients medical history, physical examination, chest x-ray & other lab tests
2 types of sample
Spot-spot
Early morning -spot
Early morning -early morning
3 important parameters to consider when coiling of sputum sample
Thickness
Evenness
Size
Size of coiled sputum smear
2 cm x 3 cm
It is used to stain bacteria that have high lipid content in their cell wall
Acid fast stain
Where does the primary stain binds in the cell wall
Mycolic acid
Color of Acid fast Bacilli & from what dye?
They are stained Red from carbol fuschin
Color of Non-Acid fast bacili and what dye is used?
Colored as blue or green from methylene blue or malachite green
Uses steam as a mordant
Ziehl-neelsen method/hot staining method
Same reagent but higher concentration of phenol w/c serve as a chemical mordant; sometimes have a wetting agent tergitol to fasten up reaction
Kinyoun’s/cold staining method
It differentiates M. Smegmatis to M. Tuberculosis
Pappenheim’s method
Differentiates m. Leprae from m. Tuberculosis
Baumgarten’s method
Selective for the cell wall of AFB
Auramine Rhodamine method
Zhiel neelsen composed of __ % phenol & __ g carbol fucshin
5% and 3 grams
Kinyoun is composed of __ g carbol fuchsin and __% phenol
4 g carbol fuchsin & 9% phenol
Positive AFB
M. Tuberculosis ATCC 25177
Positive -partially AFB
N. Asteroides ATCC 3308
Negative- non AFB
E. Coli ATCC 25922
All bacteria are non acid except:
Mycobacteria
Nocardia
Rhodococcus
Tsukamurella
Gordonia
Legionella micdadei
Parasite:
Isospora belli, Cryptosporidium & Cyclospora
It renders the cells resistant to decolorization, thus termed as acid fast
Mycolic acid
True or false: acid fastness is affected by colonial age, medium for growth & uv light
True
True or false: kinyoun’s method is ideal for Concentrated smear and partially AFB like Nocardia spp.
False: Ziehl neelsen not kinyoun’s
True or false: modified kinyoun method is not ideal for Cryptosporidia and Cyclospora parasite in stool specimen
False: it is recommended.
