- spun tube works based on centrifugal force
- lighter liquid supernatant drained off top, leaves heavier denser pellet of DNA forced to the bottom
- separated based on size, shape, density and viscosity

why does the Q -PCR graph plateau?
A
run out of materials/ components to continue building
or amplification won’t add any more to it
- no more fluorescence (increase in y axis) because no more is being made but the number of cycles (x axis increase) is still happening

ddNTP DONT have an OH group on the 3rd carbon = cannot continue the building of the chain = termination
