How do we look at xylem/phloem in plant tissue?
Under microscope and then draw what you see.
STEP 1
Scapel - cut cross section of stem.
Do this as thinly as possible .
Thin sections better for viewing under microscope.
STEP 2
Tweezers - gently place the cut sections in water until you come to use them.
This stops from drying out.
STEP 3
Add drop of water to microscope slide.
Add plant section.
Add 1/2 drops of stain eg TBO
Leave for 1 minute
STEP 4
Carefully apply cover slips so you have temporary mount.
STEP 5.
When you view specimen under microscope if youve used the stain (TBO) you should be able to see the xylem cells stained blue-green.
Phloem cells and rest of tissue appear pinkish/purple.
