4 - LIPID CHEMISTRY Flashcards
are generally insoluble in water but readily soluble in organic solvents, such as chloroform or acetone
Lipids
solubility test samples
Olive oil, Coconut oil
solubility test solvents
Dichloromethane (CHCl2), Benzene (C6H6), Ethanol (C2H5OH), Carbon tetrachloride (CCl4), Water (H2O), dilute Sodium hydroxide (NaOH) solution
Make a drop of olive oil on a piece of filter paper.
On the same filter paper, add a drop of Ethanol (C2H5OH) and a drop of Ether.
After a few minutes, note the drop made on the filter paper by holding it up to the light
Spotting Effect
positive spotting effect test
translucent spot of lipid
Samples: Olive oil, Coconut oil
To each of the test tube, add 1 mL of chloroform and shake very well.
Add dropwise Iodine (I2) solution until a yellow color is obtain.
Shake, note the number of drops of Iodine solution added on each test tube.
Test for Unsaturation
Place 1 mL of coconut oil in an evaporating dish and heat strongly.
Note the irritating disagreeable odor
Acrolein Test
This test is used for the detection of Cholesterol
Leibermann-Burchard Test
dehydrating agent in Leibermann-Burchard Test
Acetic anhydride
oxidizing agent in Leibermann-Burchard Test
Sulfuric acid
positive Leibermann-Burchard Test result
bluish-green solution
this test is used for the detection of Cholesterol
Salkowski Test
positive salkowski test result
when the Cholesterol layer becomes red or green
Green layer contains
bicholestadiene monosulfonate
Red layer contains
bicholestadiene disulfonate
