Onion root tip
(root tip because thats where meristem is)
Place root tip in stain (easier to see)
Gently squash on glass slide

Limitations of identifying Mitosis in Plant Cells

A

Difficult to distinguish between prophase and telophase
Size of cells may appear inconsistent in different specimens
Optical microscopes have low magnification and resolution so cannot observe some smaller structures
Treatment of specimens when preparing slides can damage the structure of the cells
