Q

What is the stage micrometer

A

It is a microscopic ruler on a special slide w an accurate scale, placed on the stage. The ruler is 1mm long, & divided into 100 divisions. Each division is 0.01mm or 10μm. It is used to work out the value of each of the divisions on the eyepiece graticule at a particular magnification

Q

Step by step method to calibrate microscope

A
  1. Line up the stage micrometer and eyepiece graticule whilst looking through the eyepiece
  2. Count how many divisions on the eyepiece graticule fit into one division on the micrometer scale
  3. Each division on the micrometer is 10pm, so this can be used to calibrate what one division on the eyepiece graticule is at that current magnification
