2.1.1e/f Microscope Calibrations Flashcards
1
Q
When do we use eyepiece graticule & stage micrometer
A
if you want to know the size of your specimen
2
Q
What is the eyepiece graticule
A
The eyepiece graticule is fitted onto the eyepiece. It has numbers but no units
3
Q
What is the stage micrometer
A
It is a microscopic ruler on a special slide w an accurate scale, placed on the stage. The ruler is 1mm long, & divided into 100 divisions. Each division is 0.01mm or 10μm. It is used to work out the value of each of the divisions on the eyepiece graticule at a particular magnification

Q
What does EPU stand for
A
Eye piece unit

Q
Step by step method to calibrate microscope
A
- Line up the stage micrometer and eyepiece graticule whilst looking through the eyepiece
- Count how many divisions on the eyepiece graticule fit into one division on the micrometer scale
- Each division on the micrometer is 10pm, so this can be used to calibrate what one division on the eyepiece graticule is at that current magnification
