Weeks 7-9 Flashcards
What is batch effect?
Most common confounding factor, affected by time and who performs the tests.
Stratification
Divide equally each type of subgroup between experimental groups.
Quantile normalisation
Find top rank for each patient and find mean of top 3 across samples. Bring the same level value and then use the mean.
General workflow of imaging?
Noise removal (background subtraction) Segmentation (determine what we want to capture Measurements Post analysis Machine learning
Western blot
Stimulate w insulin, buffer then centrifugate at different speeds. Fractioning. Gel electrophoresis, then use antibody that will detect the reporter.
Relative quantitation analysis
Compare abundance across samples
Absolute quantitation analysis
Number of molecules
Mass spectrometry
Light through inlet –> ion source –> mass analyser –> detector.
Continuous electrospray ionisation (ESI)
High electrical field with liquid –> spray into fine droplets –> positive charge –> can be detected.
