Light: basic, tissues Transmission: cell, cytology Scanning: 3d, surfaces Polarizing: refractive indices along different optical axes Phase: living, indices Interference: living, like phase Fluorescence – UV, fluorescent Confocal : laser, 3d
Basic tissue prep run-down:
- Fix the tissue
- Dehydrate with alcohol
- Clear by replacing alcohol with something tha can mix with wax
- Infiltrate and embed tissue with paraffin wax
- Section it in slices with microtome machine
- Stain if ya want
Basic vs Acid staining? Give some charges here:
Basic - dye is attached to a (+) charge, will color stuff with a negative charge (like DNA)
Acidic - dye attached to a (-) charge, will color stuff with a (+) charge, (like proteins)
Describe how hematoxylin/Eosin stains work:
Hematoxylin - a basic stain that will dye negatively charged stuff blue
Eosin will color all the positively charged stuff pink, because its an acidic stain
They are used together so you can see all the different cell componets

What stains do you use for fats?
oil red, sun black
Silver stains are sued to stain what?
stains the reticular fibers of connective tissue
will stain them black
