Week 12 - Mass Spectrometry Flashcards
Basic principle of Mass Spec
Measure ion acceleration and deflection in response to magnetic or electric fields
mass-to-charge ratio (m/z)
Vacuum purpose
Reduces compound fragmentation by preventing ions from colliding with air molecules
Procedure for running a non-volatile sample
Small amount of liquid placed in a probe, air locked and heated until evaporated
Should be dissolved in a solvent and allowed to evaporate
Cathode -> Anode
Formation of positive ions
Ionization Chamber
Electrons hit the sample, compound loses electrons
Parent Ion
Peak at highest mass
lower masses - fragmentation
Total Ion Chromatogram
All ions measured
Base Peak Chromatogram
Intensity of largest peak in scan
Function of time or scan #
Extracted Ion Chromatogram
One ion
Electrospray Ionization (ESI)
Liquid phase
Atmosphere Pressure Chemical Ionization (APCI)
Gas phase
Matrix-Assisted Laser Desorption Ionization (MALDI)
Solid phase
