Amp is a selective marker that allows for the selection of bacteria that carry it

unique digestive sites - that allow for insert integration, keep plasmid integrity
Non - directional cloning
Purpose to store gene X into bacteria
- Direction of insertion does NOT matter (both carry the same DNA sequence)
Directional cloning
Direction 1 - (+ strand downstream of promtoer) will express protein of interest
Direction 2 - (- strand dwonstream of promoter) will not express the protein of interest (the insert doesnt match up)
