V. Amylase Kinetics Flashcards
how to plot michaelis menten curve + equation
X-axis = substrate concentration [S]
Y-axis = absorbance [ABS]
y = ax² + bx + c
whats Km
Km (Michaelis constant): the substrate concentration at which the reaction speed is half of Vmax
whats Vmax
maximum speed of the reaction (when all enzyme is saturated with substrate)
interpretation of Km (low/high)
Low Km → enzyme has high affinity for the substrate → works well even at low substrate (not much substrate left)
High Km → enzyme has low affinity → needs a lot of substrate to work efficiently (lots of substrate left)
how to plot lineweaver burk plot + equation
X-axis = 1 / [S]
Y-axis = 1 / ABS
y = mx + c
slope formula
Km / Vmax
Vmax formula
1 / y-intercept
Km formula
- 1 / x-intercept
alternative equation of lineweaver burk
1/v = (Km/Vmax)(1/[S]) + 1/Vmax
