Use a Light microscope practical Flashcards
first step of practical
Carefully clip slide into stage
2nd step of practical
select lowest powered objective lens over the slide
3rd step of practical
Use the course adjustment to bring the stage just beneath lens
4th step of practical
Look down eye-piece lens(so you don’t damage slide) and gradually move stage downwards using the course adjustment knob. Stop when roughly in focus
5th step of the practical
To bring image into proper focus , adjust the fine adjustment knob until a clear image is obtained
6th step
To observe the image in higher magnification change to a higher objective lens readjusting the stage using course and fine adjustment knob
Why use a thin cell
So light can get through the cell so you can see the cell
Whys is the electron microscope better than light
An electron microscope has much higher magnification and
resolving power than a light microscope. This means that it can be
used to study cells in much finer detail. This has enabled biologists
to see and understand many more sub-cellular structures.
magnification
The amount that an image of something is scaled up when viewed through a microscope.
resolution
the ability of a microscope to distinguish details of a specimen or sample. In other words, the minimum distance between 2 distinct points of a specimen where they can still be seen by the observer or microscope camera as separate entities.
Safety:
Take care when handling slides and microscope
Wear gloves if using stains
Wear eye protection
Methylene blue is an irritant
