How is the sediment prepared for analysis?
Mix well, transfer 5ml to centrifuge tube
Centrifuge at 1500rpm for 5 minutes
Decant supernatant with pipette, add stain if desired
Resuspend by flicking until mixed
How is the sediment analysed under a microscope?
Low power (x10) scanning overview and allow initial quantification of elements (casts, crystals, cells)
High power (x40) allow identification of morphology and bacteria and quantify RBCs and WBCs
What cells might you expect to see in a sediment?
Erythrocytes
Leukocytes
Squamous/transitional epithelial cells
What crystals might you expect to see in a sediment?
Magnesium ammonium phosphate (Struvite) Calcium oxalate dihydrate/monohydrate Ammonium biruate Amorphous urates Bilirubin Calcium carbonate
What casts might you expect to see in a sediment?
Epithelial cell
Leukocyte
Erythrocyte
What other things might you expect to see in a sediment?
Lipids Yeast/fungi Parasites Spermatozoa Bacteria Contaminants (especially free-catch samples)
