Describe affinity chromatography
It is used to separate target proteins from a mixture.
Describe gel electrophoresis
Proteins are on a gel matrix and separate as a current is applied. Proteins are separated by charge size and shape.
Describe SDS page
Gives all molecules equally negative charge and denatured them. Separated by size only.
Describe native gel
Analyse proteins in folded state. Separated by size and charge.
What is an isoelectric point?
The pH of a protein at which it has no net charge. Protein will precipitate out of solution.
What are immunoassays?
A technique used to detect specific proteins. It can use antigens to detect antibodies.
Describe the use of antibodies to detect antigens.
The antibody specific to target antigen is bound to a plate.
Sample is added onto plate.
If antigen is present it will bind to antibody.
Label antibody specific to antigen is added which can produce a colour change.
Describe western blotting.
Used after SDS page. Separated proteins are transferred onto a solid medium.
