heat DNA between 92-98 C to break the hydrogen bonds between strands and base pairs
cool between 50-65 C to allow primer to bind to targeted sequence
heat tolerant DNA polymerase adds nucleotides to original DNA strand
heated between 70-80 C to reach optimum temp for enzymes
