How is forward grouping for A, B and O blood groups performed?
Blood is separated into red cells and plasma by centrifugation.
Red cells (expressing the antigen) are mixed with a commercial serum containing known antibody, eg anti-A.
Agglutination will occur if the antigen is present.
What is reverse grouping for A, B and O blood groups?
To ensure that results of forward typing are definitely correct.
Separated patient’s plasma is mixed with fully phenotyped ‘commercial test red cells’ that contain a known antigen, eg group A red cells.
This time agglutination will occur only is the antibody (eg anti-A) is present in the patient sample.
It is VITAL that the results from the forward and reverse typing complement each other.
