What is the pathogenesis behind paracetamol overdose?

A

Hepatotoxicity is driven by oxidative metabolites combining with sulphahydryl of hepatocyte proteins causing centrilobular necrosis. The metabolites are usually scavenged by glutathione but in large ODs this process is overwhelmed
