Topic 7: Microbial Systematics Flashcards
3 major domains
- bacteria
- archaea
- eukarya
Bacteria (domain)
- Peptidoglycan cell walls
- Most prokaryotes
Archaea (domain)
Prokaryotes that live in extreme environments but is also symbiotic with animals
Eukarya (domain)
Animal, plants, fungi
- membrane bound nucleus
- more related to archaea than bacteria
Archaea membrane phospholipids
Bipolar and form a monolayer
Archaea cell wall
Made of pseudopeptidoglycan
Serological tests
If two bacterium are evolutionarily close, antibodies by one bacteria will recognize and attach to the other
- If not, they do not react
Slide agglutination test
- serological test
- Antiserum from one bacterial strain is placed onto a slide with unknown culture
- If they are from the same serotype, the cells will clump together
Direct ELISA
Looking to see if a clinical sample has a specific microbe in it
Indirect ELISA
Looking to see if a clinical sample has a specific antibody for a particular microbe
Phage typing
Infecting the bacteria with a certain type of bacteriophage
- Only infects certain bacteria
- If a plaque is formed, the bacteria are from a certain species
