To investigate the effect of heat denaturation on enzyme action

Apparatus

Answer 1

• 2 graduated cylinders with celery
• pH buffer 7 & 1 drop washing up liquid
  (in 3 seperate water baths at 25ºC & 100ºC)

Answer 2

• Add Hydrogen peroxide to both baths
• Vol in graduated cylinders noted at 0,1&2 mins
• Rate of enzyme action calculated by 2min vol - 1min vol
• Use unboiled catalyse as control

• 25ºC graduated cylinder showed enzyme action
• 100ºC showed no enzyme action (denaturation)
