Tissue & Cell Culture Flashcards
Week 4 - two lectures
Cell Culture
Culturing of dissociated cells outside of organism and eukaryotic tissues
Use growth medium
Contact Inhibition
Cells stop dividing/growing when they come in contact with each other
Passaging
Replacing cells to avoid overcrowding
Freezing & Thawing Cells
Cryoprotectant - prevent ice crystals (DMSO)
Controlled - rapid thawing to minimize cell damage
Mycoplasma
Smallest bacteria cell
Resistant to many antibiotics
Types of Cells
Primary - normal tissue
Cell strains - limited growth/division
Cell lines - grow indefinitely
HeLa - immortalized
Induced Pluripotent Stem Cells (iPSCs)
From reprogramming animal and human differentiated cells (skin and blood)
Embryonic Stem Cells
Pluripotent cells which give rise to all somatic cell types in the embryo (understand development of specialized cells and establishment of organ structures)
