Pharmacodynamics
What is synergism?
An interaction between two or more drugs that causes the total effect of the drugs to be greater than the sum of the individual effects of each drug. A synergistic effect can be beneficial or harmful.
Types of synergism
Synergism:
- Summation or Additive
- Potentiation
What is drug antagonism?
An interaction between two or more drugs that have opposite effects on the body. Drug antagonism may block or reduce the effectiveness of one or more of the drugs.
Types of Drug Antagonism
Physiological
Chemical
Competitive
Non-competitive
Summation or additive synergism
It is a simple sum (addition) of effects, e.g., combination of agents for narcosis, that is 1 + 1 = 2
Potentiation effect of synergism
Potentiation is an interaction between two or more drugs or agents resulting in a pharmacologic response greater than the sum of individual responses to each drug or agent.
Types of competitive antagonistism
Equilibrium or reversible
Non-equilibrium or irreversible
What is competitive antagonism?
Competition between agonist & antagonist for specific site or receptor
What is non-equilibrium antagonism?
Here, the Antagonist binds to receptor with covalent bond, resulting in Irreversible blocking
