1) By membrane filtration
Filter using 0.45 micron membrane, culture in growth medium
2) Direct inoculation
Remove sample aseptically, expose to growth medium, culture and note for bacteria growth

Most Probable Number (MPN)
Dilute broth 10 fold, count CFU and multiply to estimate original amount of bacteria

Direct Transfer sterility testing
