Wright stain
Contains methylene blue, stains acidic stuff blue (DNA and RNA)
Contains eosin, stains basic stuff orange (hemoglobin, eosinophilic granules)
Prussian blue
Visualize iron granules
New methylene blue
Precipitates RNA in reticulocytes
Neutral red/brilliant cresyl green
Stain Heinz bodies
Myeloperoxidase (MPO)
Stains granulocytic cells and some monocytes
Lymph’s are negative
Sudan black B
Stains phospholipids and lipoproteins
Granulocytes and auer rods = positive
Lymph’s = negative
Specific esterase
Granulocytes = positive Monocytes = negative
Non-specific esterase
Monocytes = positive Granulocytes = negative
Periodic acid-Schiff (PAS)
Stains glycogen
Immature lymphoid, malignant erythroblasts, megakaryocytic cells = positive
Myeloblasts, normal erythrocytes = negative
Leukocyte alkaline phosphatase (LAP)
Grade 100 neutrophils 0-4+, add cells in each grade, multiply by grade and add up
Normal = 13-130
Decreased = CML, PNH
Normal = CML (in remission/infection), Hodgkin lymphoma (in remission), secondary polycythemia
Increased = neutrophilic leukemoid rxn, polycythemia Vera, CML (in blast crisis), late trimester pregnancy
