How do we identify O-H and N-H protons?

A
  • proton exchange using D2O
  • HNMR spectrum is run
  • then small amount of D2O is added and shaken
  • second spectrum run
  • any peak due to OH or NH proton disappears
  • deuterium exchanges with H present in OH and NH
