Section 3 Flashcards
DNA Replication
DNA polymerase
enzyme that copies a DNA template, catalyzes the joining of dNTP’s (add a dNTP to the 3’ hydroxyl group) . DNA poly I, DNA poly III, DNA poly (α) alpha , DNA poly (δ) delta,DNA poly (ε) epsilon, DNA poly (γ) gamma. Needs primer to synthesis DNA
DNA polymerase I
1st DNA polymerase to be identified in E.coli, mostly helps with repairing damaged DNA
DNA polymerase I
1st DNA polymerase to be identified in E.coli, mostly helps with repairing damaged DNA. REmoves RNA primers from okazaki fragments. Can act as exonuclease that hydrolize DNA or RNA.— 5’ - 3’ exonulease removes ribonucleotides from 5’ ends of Okasaki fra,, replaced by deoxynuc
DNA poly α
functions in replication of nuclear DNA in Eukaryotes
DNA poly α
FOund in a complex w/ primase and works w/ it to make short RNA-DNA fragments for Oris and during lagging strand. functions in replication of nuclear DNA in Eukaryotes
DNA poly (δ) delta
LAGGING:: Fills in gaps between okazaki fragments, MAjor function in replication of nuclear DNA in Eukaryotes.
DNA poly (ε) epsilon
LEADING:: MAjor function in replication of nuclear DNA in Eukaryotes
Replication forks
Regions of active DNA synthesis for daughter strands
DNA ligase
Joins the Okazaki fragments at their niks–forms new intact DNA strand.
DNA ligase
DNA poly δ fills at their gaps while ligase joins resulting fragments –forms new intact DNA strand.
RNase H
Removes RNA primers in eukaryotes. Enzyme that degrades RNA strand of RNA-DNA hybrids and 5’-3’ exonucleases
Sliding-clamp Proteins/(PCNA) proliferating cell nuclear antigen
Act at the replication fork. Binds to DNA poly III in Prok and DNA poly δ and ε in Euk Load the poly onto primer and maintain bound to template.. Forms ring around template DNA
CLamp loading proteins/ Replication factor C (RFC)
Loads PCNA between primer and template in rep fork uses energy from ATP hydrolysis to open clamps.
CLamp loading proteins/(RFC) Replication factor C
Loads PCNA between primer and template in rep fork uses energy from ATP hydrolysis to open clamps.
Helicase
Enzyme, unwinds parent DNA, coupled to hydrolysis of ATP
