SDS-PAGE Flashcards
1
Q
What is the difference between SDS-PAGE and native-PAGE?
A
In native-PAGE, proteins migrate differently depending on charge as well as size. In SDS-PAGE, proteins of the same size always migrate at the same rate, even if made up of different Amino Acids
2
Q
What determines the size of the pores in the gel?
A
The concentration of acrylamide
3
Q
What is the purpose of SDS?
A
It solubilises the proteins and coats them in negative charge so that they migrate towards the anode
4
Q
How are the Mass-to-Charge ratios of proteins equalised so that size becomes the only factor?
A
The proteins are denatured back to their primary structure (using heat and a reducing agent)
