Sculptra aesthetic is an injectable poly l lactic acid (PLLA) indicated for use in people with healthy immune systems as a single regimen for the correction of shallow to deep nasolabial fold contour deficiencies and other facial wrinkles in which deep dermal grid patterns (cross hatch) is appropriate
FDA approved to last over 2 years
Sculptra description
Sculptra is an injectable implant containing micro particles of poly-l-lactic acid, carboxymethylcellulose, non pyrogenic mannitol, and sterile water.
367.5 mg dose vials to be reconstituted

Massage 5-5-5
Efficacy and safety - 2 phase randomized study 0-13 months and maintenance 13-25 months
Sculptra Key Messages
“The secret to youthful looking skin”
Biostimulator - biostimulator that is biocompatible and biodegradeable
Type 1 Collagen Production - stimulates type 1 collagen production
Stimulate type 1 collagen and reinforce skins inner structure and and restore facial volume
Restores collagen gradually over time with results that can last over 2 years
How does Sculptra work?
Injected into deep dermis
Immediately the wrinkles and volume are corrected but afte r a few days the body absorbs CMC and mannitol. The PLLA particles remain in place and volume and wrinkles may appear uncorrected.
Macrophages are recruited and surround the PLLA recruiting fibroblasts
The fibroblasts deposit collagen providing structure for the skin then gradual correction takes place
Over time the PLLA particles degrade but the collagen deposits remain for long lasting improvement of wrinkles and volume defects
Sculptra vs HA filler
Sculptra increases 66.5% of type 1 collagen after 3 months - results seen at 1 month, peak month 12, and last 25 months
At 13 months 84.9% rated satisfaction as good / excellent. At 25 months 80% rated satisfaction as good / excellent
75% of 300 women preferred gradual results that last longer
HA lasts 6-12 months and is just filling lines, wrinkles, and folds
Sculptra reinforces skins inner structure and restores facial volume
Sculptra vs. Radiesse
Sculptra - 89% and 86% of patients still showed improvement at 13 and 25 months post treatment.
Sculptra is 40-63 microns and irregular / jagged particles
Radiesse - primarily a volumizer / collagen stimulation is secondary benefit. 79% of patients showed improvement at 12 months.
Radiesse is 25-40 microns and smooth particles
Sculptra components
Poly l lactic acid (PLLA), cargoC mannitol and sterile water
PLLA is sugar made from beets and corn
Sculptra is hydrophobic
Top 3 Adverse events
Swelling, tenderness, redness
Sculptra messaging
- younger than years
- beyond filling lines and wrinkles
- gradual results over time
- revitalizes collagen production
- consistent improvement over 2 years
- results last longer
- 80% good to excellent at 25 months
- subtly delivers youthful appearance
Sculptra is for younger patients, long term results, poor dermal quality, facial rejuvenation

Patients requiring facial rejuvenation
Sculptra technique
Injected into deep dermis using threading / tunneling technique
(Skin is epidermis (top), dermis (middle) subcutaneous (below dermis)
Needle in deep dermis bevel up at 30-40 degree angle
Needle then lowered to angle of 10-20 degrees and advanced forward parallel to skin surface - ensure not in vessel
A thin trail of Sculptra is applied as needle is slowly withdrawn (known as retrograde injection)
Crosshatch pattern (NLF) parallel and perpendicular to NLF .05-1cm apart
What best defines collagen?
Forms a framework to support cells and tissue
One of the most common proteins in the body
NOT - a primary component of bone structure / a vitamin supplement
Sculptra helps treat facial aging and volume loss
Fat loss
Bone loss
