Methods for screening a gene bank.
A
- Hybridisation and addition of DNA probes
- Probe can be any DNA or RNA sequence complemetary to some part of the cloned sequence
- Probe must be labeled differently to be identified bu use of radioactive isotopes or non-radioactive substance that give of colour or chemical reaction
- Cloned gene is isolated, heated (DS to SS) and cooled quickly to make probe
- It is then hybridised to denatured now SS genomic DNA
- Check hybridised molecules with autoradiography and make reference from master plate
- Enzyme activity detection method
- Immunological Screening method
- Bacterial antigens transferred from plaques to nitrocellulose discs
- Block free binding sites with bovine serum albumin
- Incubate with antiserum and wash
- Add anti-IgG/ peroxidase conjugate and wash
Add 4-chloro-1-naphthol substrate
