Required Practical 1: Enzymes Flashcards
What control set up is used for the investigation
Boiled enzymes (the enzyme has denatured so the rate of enzyme activity will be 0)
No to calculate rate of reaction using time
Rate of reaction = 1/ time
Catalase is an enzyme found in the extract of a …
Potato
When investigating the effect of substrate concentration on enzyme activity, the substrate used is…and the enzyme used is….
Hydrogen peroxide
Catalase
When the filter paper disc covered in catalase is placed at the bottom of teh hydrogen peroxide solution what causes it to rise back up to the surface
The oxygen being produced from catalase (the enzyme) hydrolysing sodium peroxide (the substrate)
Therefore we use the mean time taken for the filter paper disc to rise to the surface for each hydrogen peroxide concentration to see how the substrate concentration effects the rate of enzyme activity- a faster time means a higher rate of reaction as the substrate is hydrolysed at a faster rate to produce oxygen more quickly
Control. Armables for investigating effect of substrate (hydrogen peroxide) concentration on teh rate of enzyme activity
Volume of hydrogen peroxide
Temperature (carry out at room temperature)
Catalase concentration
Material of paper disc (same absorbance)
Surface area of paper disc
pH
Diameter of specimen tube (so that the distance from the bottom to the surface of the solution is teh same for each concentration)
Amylase hydrolyses starch into…
Maltose (not glucose)
