What is the proteome?
The proteome are all the proteins present in a cell at a particular time
How many copies will rare and common proteins have?
Rare proteins < 20,000 copies
Common up to 100 million copies
What do we use to separate proteins in one dimension?
One dimensional protein separation In poly-acrylamide gels under applied eclectic field.
What is SDS used for when separating proteins?
It denatures proteins and keeps them soluable
What is the principle of separating proteins via SDS-PAGE?
Separation based on charge:mass ratio
What does PAGE stand for eg. SDS-PAGE or 2D-PAGE
polyacrylamide gel electrophoresis
What are the safety precautions when doing SDS-PAGE?
Acrylamide is toxic and absorbed via the skin
How do we separate proteins of similar masses?
2D-PAGE
How do we do a 2D-PAGE?
Separation along a pH gradient with applied electric field.
Then use SDS-PAGE which Separates based on mass
How can we detect proteins with western blot?
Detection via specific antibodies
How can we compare two samples of cells?
Grow cells with different isotopes so they can be identified after 2D
