Recombinant DNA
DNA composed of genetic material from different sources, often different species
Recombinant DNA: Restriction Endonucleases
Enzymes that come from bacteria that cleave interior of DNA at restriction site within target sequence, creating restriction fragments
Recombinant DNA: Sequence Specificity
Each type of enzyme always cuts at same spot in same way, producing same restriction fragments
Type of Cuts: Staggered Cut
Sticky ends/overhangs: unpaired nucleotides at end of each end –> can only form base pairs with other sticky ends with complementary sequence (specific)
Type Cuts: Blunt Ends
No overlap of bases, cut at center of restriction site –> no specificity as any blunt end can combine
DNA Ligase
Joins together DNA with matching ends
