Real-Time PCR Flashcards
Understand Real-Time PCR
1
Q
What does polymerase do?
A
Polymerase synthesizes a complementary sequence of bases to any single strand of DNA providing it has a double stranded starting point.
2
Q
What are primers?
A
Primers are pieces of DNA complimentary to your gene of interest. They allow you to choose which gene the polymerase will amplify. They are known as primers because they prime the DNA sample ready for the polymerase to bind and begin copying the gene of interest.
3
Q
What is the optimal temperature for polymerase to operate?
A
72 Celcius
4
Q
What temperature is the reaction started at to allow double stranded DNA become single stranded?
A
95 Celcius
5
Q
What temperature allows primers to bind the the gene of interest?
A
60 Celcius
