Darkfield Microscopy
Used to view living, unstained microorganisms. Utilizes a darkfield condenser containing an opaque disc, only allowing light in that has been refracted by the specimen into the objective lens, which forms an image.
Used when analyzing thin and distinctly shaped bacteria
Phase-Contrast Microscopy
Optimal for viewing living cells, examining bacterial motility and intracellular structures.
Exploits differences in refractive index between the cell cytoplasm and surrounding medium. A phase plate shifts light reflecting off of cells to form a sharp image of cells darker than the background
Microscope Condenser
Gathers light waves from the microscope light source, directs them into a cone of light that illuminates the specimen
Immersion Oil Function
Increases the resolution of a microscope by replacing the air gap between the immersion reflective lens and cover glass with a high refractive index medium (reducing light refration and leading to a more crisp picture)
Relationship Between Amount of Light Needed and Magnification
The greater the magnification, the more light that is required
Discerning True Motility from Brownian Movement
True motility will demonstrate actual movement of the cell, while a cell does not move in Brownian movement

Viewing under electron microscope to observe the rotation of the flagella which causes the cells to either move forward or reverse
