MOA: Block ET receptors –> vasodilation

Contraindication:
– Pregnancy (BBW)
Eicosanoids
Example: PGI2 analogs “prosts” and PGI2
agonists: Epoprostenol
MOA: Stimulate IP receptors on vascular SM
–> vasodilation
AEs: Hypotension, Headache
– GI adverse effects
– Jaw pain, pain in extremities
CIs: N/A
NO and other sGC activators
Example: NO and Riociguat
MOA: Activates sGC in VSM –> increases cGMP
–> vasodilation
AEs: Hypotension, Headache
CIs: N/A
PDE5 inhibitors
Example: Sildenafil
MOA: Inhibit PDE5 in vascular SM –> decreases degradation of cGMP –> vasodilation
AEs: Hypotension, Headache
CIs: Patients taking organic nitrates
