Protein Sequencing (Dr.Miles TAMU) Flashcards
Primary Structure
is the linear sequence of amino acids
Secondary Structure
the spatial conformation of the polypeptide’s backbone without regard to the conformation of the side chains. Includes α−Helix, β−Sheets and β−Turns.
Tertiary Structure
refers to the three dimensional structure of the entire polypeptide including the side chains
Quaternary Structure
many proteins are composed of 2 or subunits. The quaternary structure is
the spatial arrangement of these subunits
What was the first peptide to have it’s primary structure determined
Insulin
Trypsin
Digestive enzyme that will cleave at the C-terminal side of lysine(k) and Arginie(R) unless followed by a proline
Elastase
cleaves Gly(G), Ala(A), Serine(S), and Val(V) unless followed by proline
Chymotrypsin
Cleaves Phe(F), Trp(W), and Tyrosine(Y) unless followed by proline
Cyanogen Bromide
Will cleave Methionine (M)
