Creutzfeldt Jacob Disease Biochemical Basis
Involves interaction between PrP^Sc (form of prion protein that causes disease) and normal membrane protein PrP^c
- PrP^Sc has identical 1ary structure but higher proportion of beta-pleated sheet
- When solute PrP^c comes into contact with PrP^Sc it causes PrP^c to acquire abnormal PrP^Sc structure
- Forms soluble aggregates of protein
How do molecular chaperones assist folding?
Bind to short segments of protein to facilitate correct folding
How do chaperonins assist folding?
Form folding chambers which provide stable environment to encourage correct folding
