Principles Flashcards
What is bichromatic blanking
Measuring the absorbance of a blank
Measuring absorbance at 2 wavelengths to provide a blank reference point
What light source is used in a spectrophotometer
Continuous spectrum
Colorimetric assay
The use of a spectrophotometer to determine the concentration of a coloured compound in solute
Read absorbance of sample
Beer Lamberts law
Nephelometric
Used to detect the concentration of proteins in serum
Based on the concept that particles in solution will scatter light passing through the solution rather than absorbing the light
Photometric
To measure the intensity of light transmitted through a sample by converting light into electricity
Only uses light only in the visible range
Turbidimetric
The measurement of turbidity by quanitfying the degree of ‘attenuation’ of a beam of light of known initial intensity
End point
An enzyme assay that is run for a predetermined amount of time
The amount of substrate consumed and/or the quantity of product formed is measured over the coruse of the reaction
Used to determine the rate of a reaction
Kinetic
The progress of the reaction is continuously measured as substrates are converted into products
Measure at multiple time intervals to produce a graph of absorbance vs time
