Pre-module stuff (Important) Flashcards
What are the 3 main components of DNA?
- Sugar
- Base
- Phosphate backbone
What bonds form between the bases in DNA?
Hydrogen
What bonds form between the sugars in DNA?
Phosphate bonds - this forms the sugar phosphate backbone
In what way are DNA strands arranged with each other?
In an antiparallel direction from the 5’ to the 3’ end
What is the minor groove in DNA?
Where the sugar phosphate backbones are far apart
What is the major groove in DNA?
Takes place where the sugar phosphate backbones are close together
What are the components of a base unit?
- Pentose sugar
- Phosphate
- Purine or pyramidine base (nitrogenous bases)
YOU MUST LEARN HOW TO DRAW A BASE
GO TO THE SLIDES
What is the difference between the purines & the pyrmidines?
Purines have a 2 ringed structure whereas pyrimidines have only 1 ring which the bonds form on
What are the Purines?
Adenine & Guanine
What are the Pyrimidines?
Cytosine, Thymine (DNA) & Uracil (RNA)
What is the difference between ribose & deoxyribose?
Deoxyribose has no hydroxyl bond
(Bottom right bond has H rather than OH)
What is a nucleoSide vs a nucleoTide?
Nucleoside = sugar & base
Nucleotide = sugar, base & phosphate
Which base pairs match up with each other?
A –> T (U)
G –> C
(Complementary base pairing)
What are the 3 forms of DNA?
A = right helix
B = right
Z = left
What are the characteristics of A DNA?
- Right helix
- 11 base pairs per turn
- 2.6 (Å) = distance between base pairs
- 23 (Å) = diameter of helix
What are the characteristics of B DNA?
- Right helix
- 10 base pairs per turn
- 3.4 (Å) = distance between base pairs
- 19 (Å) = diameter of helix
What are the characteristics of Z DNA?
- Left helix
- 12 base pairs per turn
- 3.7 (Å) = distance between base pairs
- 18 (Å) = diameter of helix
Describe the role of B DNA?
- Common type od DNA found in cells
- Large major groove
- Some protgeins can ‘read’ the base seq in the major groove
What is the nucleosome?
The subunit of chromatin
What is chromatin
What is the protein start codon base sequence?
ATG
What are the main componenets of a eukaryotic gene?
- RNA starts
- Protein stat codon (ATG)
- Introns (not in mRNA) & exons (in mRNA)
- Protein stop codon (TAG, TAA, TGA)
- RNA end
What is the protein stop codon base sequence?
TAG, TAA, TGA
Got to slide 13 promoter regions
