Post PCR- various instruments and HCV genotyping Flashcards
An automated extraction, amplification, detection system used for HIV viral loads, HCV viral loads, and HBV viral loads.
Ampliprep/TaqMan (Roche)
Completely automates sample prep by performing the complex steps of DNA or RNA extraction in its advanced microfluidic cartridges. Currently running Beta strep group B on mothers, Enterovirus on CSF, and Mycobacterium tuberculosis PCR.
GeneXpert system
Fully automated sample processing uses a combination of lytic enzymes and extraction reagents to perform cell lysis, and nucleic acid extraction. Detection is based on multi-wavelength fluorescence detection. Currently the laboratory performs MRSA and clostridium difficile PCR on this instrument.
BD Max
Based on flow cell fluorometry with ____- developed innovations. The fluidics, optics, robotics, temperature control, software, and xMAP microspheres work together to enable simultaneous analysis of up to 100 analytes in a single test sample.
Luminex 200
Currently using this platform for a respiratory viral panel which simultaneously detects 9 viruses and for a GI pathogen panel that detects 11 bacterial, viral, and parasite pathogens.
Luminex 200
An automated GenProbe system used for detection of Chlamydia trachomatis (CT), Neisseria gonorrhea(NG), and Trichnomonas vaginalis.
Tigris
The system uses target capture, transcription mediated amplification, hybridization and dual kinetic detection to simultaneously detected CT and NG
Tigris
The centrifugal rotary design of the _______ makes it a precise and versatile real time PCR cycler. Currently, running different type of mutation assays on this platform.
Quiagen Rotor Gene Q
A fluorescence-based capillary electrophoresis system that contains 16 capillaries. Using it for fragment analysis applications, HIV-1 genotyping and IGH/TCR gene rearrangement on this instrument.
ABI 3130
A real time PCR instrument on which focus diagnostics laboratories worked closely with 3M to create user friendly software customized by medical technologists. Allows all DNA assays and RNA assay to be run simultaneously. Performing viral load testing for adenovirus and BK virus and detection of Bordetella pertussis/parapertussis and group A strep on this instrument.
3M integrated cycler
This technology helps solve complex RBC and platelet compatibility cases. It utilizes the genetic information of blood donors and patients to predict RBC, platelet, and leukocyte phenotypes. Assay is not influenced by immunoglobulin coating of the RBC, recently transfused RBC or any form of polyagglutination.
Immucor’s BioArray Molecular BeadChip
Similar to Capillary electrophoresis sanger sequencing in that the bases of small fragments of DNA are identified from signals emitted as each fragment is re-synthesized from a DNA template strand. ___ _______ ____ extends this process across millions of reaction in a massively parallel fashion, rather than being limited to a single or few DNA fragments.
This is Illumina MiSeq; Next generation sequencing (Blank)
Why is it important to know the HCV genotype of a patient
Because many patients infected with HCV remain asymptomatic for decades but eventually develop chronic hepatitis which may result in liver cirrhosis or to liver cancer
Name and describe the type of assay used to determine the HCV genotype
The versant HCV genotype 2.0 assay is a line probe assay, LiPA, intended for use on serum or plasma samples
Discuss the type of probe used on the nitrocellulose strip in the Versant HCV genotype 2.0 assay
genotype-specific DNA probes
