PK/PD of Infection Flashcards
1
Q
PK Aspects
A
- Dosage regimens
- Concentration versus time in serum
- Absorption, distribution, elimination
- EX: time-kill curves
2
Q
PD Aspects
A
- Concentration versus time in tissues and other body fluids ==> Pharmacologic or toxicologic effect
- Concentration versus time at site of infection ==> antimicrobial effect versus time
3
Q
PK/PD Parameters
A
Combine information on what the body does to the drug as well as what the drug does to the body/bacteria
4
Q
3 Antibiotic Groups
A
- Time > MIC: B-lactams, Linezolid
- AUC/MIC: Macrolides, tetracyclines
- Peak: MIC: Fluoroquinolones, aminoglycosides, metronidazole, daptomycin
5
Q
Aminoglycosides + PK/PD
A
- Allows to select dosing regimen that minimizes toxicity from aminoglycoside therapy
- Single daily dose is preferred (some exceptions like pregnant women and impaired renal fxn)
- Toxicity is both time and concentration dependent
- Aminoglycosides shown to have PAE
- Used for Gram “-“ infections
6
Q
PAE
A
- Post-antibiotic effect
- Persistent suppression of bacterial growth after the course of therapy has ended
7
Q
AUC
A
Area under curve
8
Q
MIC
A
Minimal inhibitory concentration
