Pharmacometrics/TDM Flashcards
Importance of pharmacometrics
- Safety analysis
- Efficacy analysis
- Individualised dosage specimen
- Rational drug treatment
- Drug response predictability
- Drug development
Pharmacometrics models
- PK
- PD
- PK-PD, aka exposure-response
- Physiologically-based PK
- Disease progression model
- Trial model
Steady state plasma concentration
This is the point at which a repeatedly administered drug that accumulates in the body has a rate of elimination that equals the dosing rate or the amount administered per time.
Loading dose
A single or few quickly administered doses given in the beginning to attain target conc rapidly.
Maintenance dose
Repeatedly administered dose given at specific intervals after attaining target steady state to maintain same by balancing elimination.
Drugs administered with loading and maintenance dose
Digoxin
Chloroquine
Amiodarone
Drugs commonly monitored with TDM
Digoxin
Phenytoin
Carbamazepine
Gentamicin
Phenobarbital
Cyclosporine
Propranolol
