what drug class is buspirone?
5HT partial receptor agonist
what is buspirone used to treat?
generalised anxiety
what is buspirone’s benefits?
it does not cause sedation, tolerance, physical dependance, anticonvulsant or muscle relaxant effects.
what is buspirone’s mechanism of action?
it acts as an agonist for 5HT(1A) autoreceptors present on presynaptic neurones in the dorsal raphe and on the postsynaptic neurones on the hippocampus.
it also acts as an antagonist for D2 autoreceptor so more dopamine is released.
what is the net result of buspirone?
serotonergic activity is suppressed while noradreneergic and dopaminergic cell firing is enhanced.
what drug class is diazepam?
a benzodiazepine
what is diazepam’s mechanism of action?
it binds to an allosteric site on benzodiazepine receptors which is a GABA-A receptor. this leads to enhancing GABAs effects which allows chloride channels to open, which causes hyper polarisation and prevents further cell excitation
what do benzodiazepine receptors mediate?
sleep muscle relaxation anticonvulsant activity motor coordination memory
what is diazepam used to treat?
anxiety and as a muscle relaxant
what drug class is donepezil?
a reversible acetylcholinesterase inhibitor
what is donepezil used to treat?
dementia
it used to be used for alzheimers
what is donepezil’s mechanism of action?
it reversibly inhibits brain acetylcholinesterase, leading to an increase in ACh at cholinergic synapses, enhancing cholinergic function
what drug class is memantine?
a NMDA receptor antagonist
what is memantine used for?
dementia
what is memantine’s mechanism of action?
the blockade of current flow through channels of NMDA receptors–a glutamate receptor
