What is the general MoA, contraindications and side effects of phosphodiesterase 5 inhibitors?
A
= ^ cGMP = smooth muscle relaxation of the blood vessels of the corpus cavernosum
Sildenafil (short-acting, take 1 hr before sex), Tadalafil (longer-acting, can take once daily)
Contraindications: being on other vasodilator drugs (nicorandil [K+ channel activator] or nitrates), hypotension, stroke or MI within the last 6 months
Side effects of sildenafil / tadalafil: blue-coloured vision, nasal congestion, flushing
