Pharmacokinetics Flashcards
Bioavailability factors
route of administration, drug enters the systemic circulation, conc appearing in the plasma is entered over time to say area under the curve
Extraction or extraction ratio
fraction or percentage of drugs being removed from the perfusing blood during its passage through the organ
If it’s high, bioavailability is low
dosage regimen
plan for drug administration so that therapeutic levels remain in the blood without exceeding the minimum toxic concentration
ideally used with knowledge of both therapeutic conc and minimum toxic concentrations
Maintenance Dosage:
Because the maintenance rate=rate of elimination at a steady state
Maintenance Dosage
bioavailability
Loading Dose(if therapeutic concentration must be given rapidly)
Vd multiplied with desired concentration divided by bioavailability
therapeutic window
safe range between between minimum therapeutic concentration and minimum toxic concentration
theophylline
therapeutic concentration range of 8-20 mg/L but maybe toxic at concentration of more than 15-20 mg/L therapeutic window=8-16 mg/L
Corrected Dosage
100 mL/min
In elderly patients
72xScr
